Fluorescence spectroscopy contributes significantly in the fields of biological analysis, material characterization, and surface science. Especially, to uncover heterogeneities encoded in ensemble experiment, single-molecule (SM) analysis has attracted considerable attention in multidisciplinary realms. When an emitter is placed in close proximity to the plasmonic nanostructure, the photostability, spontaneous emission rate, and intensity are modified due to the increased local density of states (LDOS).<br/>Recently, a novel spectroscopic methodology has been invented, which is named shell-isolated nanoparticle-enhanced Raman scattering (SHINERS). The shell-isolated nanoparticle (SHIN) is prepared by coating an ultra-thin dielectric shell onto the metal core, which could spread as “plasmonic dust”. In virtue of Ag SHINs, we have obtained an ensemble fluorescence enhancement up to 40,000-fold and a Purcell factor around 280. In the presence of shell-isolated nanoparticle platform, the SM emission spectrum with high signal-noise ratio (SNR) is achieved to explore the transient conformational variation. We are optimistic Ag SHINs will path an avenue to understand light-mater interaction in the molecular level.
