Examining the collective impact of combined parameters is crucial for systemically understanding the interactions between nanoparticles and biological systems and will be helpful for design of new nonoparticles with better safety profiles. To systemically investigate the effect of nanoparticles' physicochemical properties in their biological effects, we designed and synthesized 6 combinatorial nanoparticle libraries including 3 core materials (Pt, Au, and Pd), 2 particle sizes (5 and 25 nm) and different surface chemistry. We studied the cellular uptake, cyototoxicity and reactive oxygen species (ROS) generation after incubation with different nanopartiles and correlated the results with their physicochemical properties. Our results showed that for nanoparticles with same core materials, nano-hydrophobicity was directly correlated with the cellular uptake of nanoparticles, regardless of particle sizes. Increased ROS generation was found with increased cellular uptake. However, nanoparticles with different core materials showed different profiles in terms of cellular uptake, ROS generation, and cytotoxicity, showing the effects of core materials. Specifically, as a result of different redox activities of nanoparticles, with same surface chemistry and size but different core materials, different ROS generation can be found. The results reported here indicate the importance of the interplay among core materials, sizes and surface chemistry on directing nanoparticles' biological effects, and offer insight into engineering nanoparticles with predictable biological effects.
