The accurate quantification of metal ions in biological samples can be challenging. Inductively Coupled Plasma Mass Spectrometry (ICP-MS) is emerging as the gold standard for these studies; however, difficult sample matrixes, dynamic equilibrium, and overlapping instrumental interferences present challenges for many samples. We are developing methods to quantify total metal ions in biological samples as well as to determine metal ion speciation by coupling liquid chromatography to ICP-MS (LC-ICP-MS). In one area, we have developed LC-ICP-MS to measure iron speciation in blood plasma of healthy volunteers who have been administered the iron nanoparticle drug, Ferrlecit (sodium ferric gluconate). Our method allows us to separate and quantify the iron nanoparticle, iron proteins (transferrin, ferritin, albumin) and labile or free iron (iron citrate). In the second area, we are characterizing protein stabilized gold nanoclusters (AuNCs). AuNCs are sub-nanometer sized particles that are composed of between 5-25 gold atoms cloaked by a protein. AuNCs exhibit distinct fluorescence emission properties and show promise as sensors for cellular processes, new imaging agents and real-time chem-bio sensors. Not all AuNCs retain protein activity, and the factors that control activity are not understood. Our efforts to identify the factors that modulate protein AuNC formation using ICP-MS and LC-ICP-MS will be presented.
