Hydrogen-deuterium exchange (HDX) monitored via mass spectrometry has established itself as a concrete approach for studying many mechanistic theories behind various reactions. Deuterium (D)-labeled compounds have become an interest to many scientific fields, as they can be used to study reaction mechanisms, biosynthetic pathways, reaction selectivity, and metabolic stability of drugs etc.<br/>Gas Chromatography-Mass Spectrometry (GC-MS) analytical assay of HDX was successfully developed. A series of reactions involving HDX on the benzylic position of Xylene thus are performed using a deuterium-enriched material, deuterium oxide (D2O) under palladium-on-carbon (Pd/C)-catalyzed conditions.
