Analysis employing multiple techniques can often provide complementary understanding in helping identify minor components present in compound mixtures. We have used NMR and LCMS techniques to identify for the first time Δ-8-tetrahydrocannabinolic acid (Δ- 8-THCA) as a minor isomerization product of the naturally occurring isomer Δ-9- tetrahydrocannabinolic acid (Δ-9-THCA). The isomerization product was identified as a minor component from a Pd-catalyzed hydrogenation of pure crystalline Δ-9-THCA to prepare hexahydrocannabinolic acid (HHCA).NMR and LCMS data to support the identification of this isomerization product of Δ-9-THCA will be presented. The identification and access to this carboxylated acid precursor of the widely known Δ-8-THC isomer of Δ-9-THC may prove useful in developing cannabinoid based medicines for the treatment of medical conditions underpinned by the endocannabinoid system.
