Combinatorial approaches in directed evolution were proven to be more efficient for optimizing reaction conditions, in particularly for discovering new methodologies in organic synthesis and early drug discovery programs. Here, we presented the series of new methodologies which were discovered and optimized via combinatorial routes. Those methods provided 1. novel syntheses of (z)-β-amino α-phenoxy-α,β-unsaturated amides via rearrangement and replacement reactions from formamidines and α-phenoxy acide chlorides. 2. facile synthesis of poly-heterocyclic compounds from corresponding formamidines and amino-esters. 3. Regio-specific and library friendly synthesis of N-1-alkyl-2-unsubstituted benzimidazoles. 4. Novel synthesis of benzimidazoles vis palladium catalyzed Suzuki coupling and cyclizing reactions in one pot. Such new methodologies have been enabled us to access many kinds of compounds in efficient ways, such as selective synthesis of (z)-β-dimethyl amino-α,β-unsaturated amides (A); poly-heterocyclic compounds (B); N-1-alkyl-2-unsubstituted benzimidazoles (C); variety of benzimidazoles (D) and benzo[1,4]oxazine-2-carboxamide (E) -Scheme 1.
