YOYO-1 is a commonly used DNA staining dye that is fluorescently bright in DNA but very dim in water. The major accepted assumption of its nonradiative decay pathway is that the photo-excited electrons thermally relax via the rotation at a bridging methine that connects the two moieties of the molecule. We use two dimension correlation analysis and kinetic analysis of ultrafast transient absorption spectroscopy data to support the intermolecular charge transfer mechanism for the nonradiative relaxations, which explains its fluorescent quenching mechanism in different environments.
