The formation of coordination compounds with platinum central metals such as cisplatin or carboplatin had a huge impact on current cancer chemotherapy. However, the gamut of cancers that can be treated with platinum compounds is narrow and treatment efficacy are limited from side effects and resistance phenomena. Hence, new drugs with fewer severe side effects, reduced tumour resistance and different mechanisms of action are needed. In addition, the development of anticancer agents that are effective against many different tumours is imperative due to the staggering number of new diagnosed cases of cancer every year. Among the many early and late transition metal-based potential anticancer drugs, ruthenium (Ru) compounds have garnered a lot of interest. For example, the Ru(III) compounds NAMI-A and KP1019 are in Phase II clinical trials and several organometallic and luminescent polypyridine Ru(II) compounds have displayed high anticancer activities against several cancer cell lines. Since most of the polypyridine Ru(II) antitumor compounds contain N-donor neutral ligands, it was decided to introduce anionic N^O donor bidentate ligands into the coordination sphere. We have synthesized six (6) monocationic Ru(II) complexes with general formula [Ru(N^N)2(N^O)]+; where N^N = bpy, phen and dpq and N^O = 2-hydroxy-6-methylpyridine or 2-chloro-6-hydroxypyridine, which were characterized by ESI-MS, electronic absorption spectroscopy and single crystal X-ray diffraction. Their anticancer properties were assessed against HELA and OVCAR-8 cancer cells and compared with that of cisplatin.
