KP1019 is a ruthenium-based anticancer drug that effectively induces apoptosis in cancer cells. The mechanism of the drug has not been determined. It has been suggested by some studies that KP1019 binds DNA, but no studies have looked at the interaction of KP1019 with DNA in the cell; therefore, this study analyzes the function of KP1019 in paired healthy and cancerous epithelial tissue. Telomeres serve as a biomarker for DNA damage because they are the first part of the genome to be damaged by chemotherapeutic agents that mutate DNA. Thus, if the cell free telomere concentration in the cell media increases, then the drug damages DNA. The data collected indicates that both total cell free DNA and cell free telomeric fragments increased after treatment with KP1019 relative to the untreated controls; therefore, we have concluded that KP1019 induces apoptosis via DNA damage.
