An electrochemical sensor based on an arginine (Arg) functionalized graphene (Arg-G) nanocomposite was fabricated for sensitive detection of Acetaminophen (AP). The nanocomposite was characterized by transmission electron microscopy, Raman spectra, etc. The electrochemical behaviors of AP on the Arg-G composite film modified glassy carbon electrode were investigated by cyclic voltammetry, quartz crystal microbalance, and differential pulse voltammetry. The interaction between Arg and AP was also investigated by Fourier Transform Infrared Spectroscopy, UV Spectroscopy, and fluorescence spectra. The results indicated that Arg can form hydrogen bonds with AP, which could enhance the electrochemical response of AP. This fabricated sensor displayed excellent analytical performance for AP detection over a range from 0.1 to 3000 μmol L<sup>−1</sup> with a detection limit of 0.05 μmol L<sup>–1</sup> (S/N=3). The sensor also showed good performance for AP detection in serum. Moreover, the proposed electrochemical sensor also exhibited good reproducibility and stability and has been used to detect AP in tablets with satisfactory results.
