Monitoring concentrations of a variety of drugs in biofluids to determine their efficacy and toxicity requires high-throughput screening methods. Organosiloxane (OSX) polymers prepared by sol-gel chemistry were developed as sample supports and electrospray ionization (ESI) emitters in a single material for use in ambient ionization mass spectrometry for the rapid analysis of drugs in biofluids.  By using sol-gel chemistry, the polarity and porosity of the OSX polymers can be controlled.  Small volumes (1-2 mL) of drug samples in urine were deposited on an OSX polymer, allowed to dry, then electrosprayed from the polymer tip into the mass spectrometer without sample pretreatment. The utility of OSX polymers as substitutes for paper in polymer-spray mass spectrometry is demonstrated using a variety of drugs of different polarities, including narcotic drugs and antibiotics.<br/>
