The ability to respond to external signals is a fundamental property of all living organisms. Signaling is often controlled by reversible phosphorylation of proteins, catalyzed by &gt;500 protein kinases and &gt;140 protein phosphatases.  Aberrant signaling mediated by these enzymes can lead to numerous diseases and thus kinases are now well established as therapeutic targets while targeting phosphatases is an emerging area.  Importantly, molecular methods are necessary to (a) understand the polypharmacology of small molecule kinase inhibitors and (b) deconvolute the specific activity of any desired kinase or phosphatase within living cells in order to probe their function.  Progress towards these dual goals will be presented using multiple protein engineering approaches, where small molecules control the function of appropriately engineered protein kinases and phosphatases in living cells.<br/>
