The Williamson lab is exploring the mechanism of auto-reduction of high-valent iron species in hemoglobin, with the intent to uncover the features that regulate unwanted protein oxidative damage. This mechanism is important biologically, because the unwanted auto-reduction of high-valent hemoglobin is associated with heart disease. We probe the kinetic and thermodynamic parameters of the mechanism utilizing UV-visible transient absorption spectroscopy. By analyzing the results of this technique at varying temperatures and pHs, we can determine if there are different features that are important in regulating the mechanism at physiologically low pH (inflammation sites) and neutral pH (blood based environments).
