Cucurbituril based host-guest systems have attracted much attention for the controlled release of drugs because of their ultratight and selective binding with guests, water solubility, and low toxicity. In this work, we discuss the synthesis of an autonomous DNA transducer based on Cucurbit[7]uril (CB7) and adamantane linked DNA chimeras derived from a split ATP binding DNA aptamer. The synthetic transducer can convert an ATP input signal into functional displacement of a CB7 guest (an inhibitor of carbonic anhydrase II) from the CB7 host via a combination of ATP induced duplex formation and intramolecular host-guest interactions. This strategy that combines host-guest supramolecular chemistry and nucleic acid structure switching is expected to be broadly applicable for the development of input-responsive drug release technologies since the DNA aptamer architecture can be adapted to a variety of alternative biomolecules.
