Parthenolide, a natural product found in many plants such as feverfew and magnolia grandiflora, inhibits several cellular pathways and has provided inspiration for new chemotherapeutic agents. Parthenolide impacts the heat shock response system through the Hsp70/Hsp90 chaperone complex and has been shown to covalently adduct Hsp70. Cysteine to serine mutants of Hsp70 and Hsp90 have been generated to assess the effect of parthenolide on mutant function. Parthenolide inhibits the ATP-binding ability of Hsp70 and also affects the ATPase activity as well. An alkyne-bearing derivative of parthenolide has also been used to fluorescently track protein adduction in human leukemia cells.
