Rhodamine derivatives have been used as a range of molecular sensors and some of these derivatives have shown efficacy for intracellular monitoring. Rhodamine can be present in an open form that is colored and fluorescent and a closed form that is non-colored and non-fluorescent. In its open form, there is an sp<sup>2</sup>, sp<sup>2 </sup>single bond that has limited rotation. Much is known about the formation of rhodamine derivatives but little attention has been paid to forming asymmetric versions of rhodamine. Asymmetric derivatives of rhodamine are chiral compounds that are atropisomeric in the open form. These atropisomeric derivatives have the potential to function as switches or fluorescent sensors for chiral compounds. Presented here is an accessible path to form a range of asymmetric rhodamine derivatives. The synthesis, resolution, and preliminary testing of these asymmetric rhodamine derivatives will be presented.
