Multicomponent reactions (MCRs), like the Ugi-Smiles coupling, can provide efficient preparation of diverse libraries of complex biologically-active scaffolds. The Ugi-Smiles reaction couples phenol, amine, aldehyde, and isocyanide components in a one-pot reaction to form a highly functionalized amide product. Use of propargylamine as a reaction component provides the opportunity to exploit the efficiency of the Ugi-Smiles coupling, by combination with an azide to produce a potentially biologically active triazole. Current work is focused on optimization of the reaction conditions, exploration of component substitution, and reduction of the nitro substituent in the triazole product.
