A reliable and concise synthesis of protected 5-cyano-L-tryptophan has been developed to enable infrared spectroscopic investigations. In order to assess the potential of differently substituted cyano-tryptophans, several model cyano indole systems were characterized. The FTIR spectrum at different substitutions were measured and absorption cross sections were determined. A short model peptide of Trp<sub>5CN</sub>–Gly–Phe<sub>4CN</sub> was synthesized to measure vibrational coupling between differentially labeled side chains via 2D IR spectrosocopy. The 2D IR spectrum was simulated to uncover the vibrational coupling between the labels.
