C-terminal diversification is a powerful transformation for accessing synthetic peptide targets. Incorporation of Fmoc-o-amino(methyl)aniline (MeDbz) as a linkage between the solid support and peptide can assist with functionalizing C-terminal peptides following solid phase peptide synthesis (SPPS). Displacement of the activated <i>o</i>-amino(methyl)aniline (MeNbz) linker with a variety of amino acids (i.e. Gly, Ala, Ile, and Pro) allows access to C-terminal point mutations. Although C-terminal manipulations are traditionally hindered by epimerization, our method has minimized this complication. Thus, demonstrating a convergent method to access C-terminally elongated peptides.
