Biomarker-based immunoassays are specific and quantitative; however, they depend on intradermal or subcutaneous extraction of fluid. One of the hurdles to extraction of interstitial fluid (ISF) is the stratum corneum, the uppermost layer of “dead skin.” A method to bypass the stratum corneum and extract ISF would be an invaluable tool for the development of real-time biochemical monitors. Herein the design, fabrication and evaluation of a nanocellulose-composite microneedle array to extract ISF is presented. The superhydrophilic nanocellulose-composite, in conjunction will capillary force, enables the extraction of ISF. The nanocellulose-composite microneedles are mechanically robust and biocompatible, which will support long-term use. The proposed platform represents the first step towards the realization of a long-term wearable microsystem for analysis and profiling of ISF.
