Macrocycles are becoming important class of compounds in areas such as drug development and supramolecular chemistry, 1,5-Disubstituted tetrazole are bioisosteres to amide groups and its derivatives have attracted much attention because of their unique structure and applications as antiallergic, antihypertensive, antibiotic and anticonvulsant agents. A synthetic platform for the fast and efficient access to hundreds of macrocyclic scaffolds using multicomponent reaction chemistry (MCR) is presented. In particular the Ugi reaction may be used for the synthesis of precursors as well as the macrocyclization itself. The Dömling group has developed methods for the synthesis of α-isocyano-ω-carboxylic acids, α-amino-ω-carboxylic acids and α-amino-ω-isocyanide and its use in in macrocycle synthesis via MCR. The synthetic chemistry will be discussed and as well as an application of these artificial macrocycles for the discovery of potent p53-MDM2 antagonists.
