Neurodegenerative processes are responsible for the development of Alzheimer’s and Parkinson’s disease, among others, leading to billions of dollars in disease burden to our society. As a result, there is significant interest in elucidating specific mechanisms of both neurotoxicity and neuroprotection. The limonoids are one class of natural products that show significant neuroprotective activity against glutamate toxicity. We are investigating a strategy for the synthesis of furanolactone limonoids through an early-stage catalytic arylation reaction under development in our group. This strategy should be amenable to the synthesis of analogs and probe molecules for the investigation of neuroprotective properties.
