Glycosaminoglycans (GAGs), especially heparin and heparan sulfate (HS) have been recognized to be playing critical role in many biological processes including cancer, neurological diseases and inflammations. However, accessing to well defined and therapeutically useful natural heparin/HS fragments has been a challenge. Thus, there is a great demand for stable, controllable, and well-defined synthetic alternatives of heparin/HS fragments with more effective biological functions. PharmaRen Discovery LLC has been at the forefront of the efforts to bring important building blocks to the market and make them available to the scientific community in mg-kg quantities. The first generation of the building blocks are now available through a dedicated website (heparinbuildingblocks.com). In this Poster, we present the syntheses and some applications of second generation of these heparinic building blocks. The second generation of the building blocks are made up of less abundant and infrequent synthetic heparinic building blocks. These building blocks are aimed to be used in the construction of heparinoids of sporadic but well-defined configuration. They are essential in determining structure-reactivity relationships and defining the interaction between HS and HS-active ligands. The following scheme demonstrates how our new building blocks can be utilized to generate a rare heparinoid.
