Opiate addiction during pregnancy is a very important public health concern causing great dangers to both the mother and the child in utero. This illuminates the need for maintenance programs, like buprenorphine treatment, to reduce illicit drug use during pregnancy. Buprenorphine is a safe treatment option used during pregnancy which acts as a partial agonist for opioid receptors. Drug treatment during pregnancy is drastically different than treatment for the average human being because a pregnant women’s metabolism increases considerably over the nine-month period. This generates an increased focus upon the study and analysis of buprenorphine metabolite concentrations during pregnancy. This in-vitro study will be performed using cytochrome P450, the enzyme responsible for metabolizing buprenorphine in the human body.<br/>Apigenin, imperatorin, and bergamottin are three polyphenols shown to inhibit cytochrome P450. These polyphenols are found in fruits, vegetables, and juices making them easily accessible. The question we are interested in is whether dietary supplementation of polyphenols can serve as a possible inhibitor of cytochrome 450 metabolism of buprenorphine and extend the effect of the drug on pregnant women using it. Buprenorphine, cytochrome P450, and the individual polyphenols will be combined and analyzed through High Performance Liquid Chromatography. The analysis will be used to determine any change in buprenorphine metabolite concentration in the presence of cytochrome P450 accompanied with the natural, phenolic compounds.
