Sample processing protocols were developed for liquid chromatography mass spectrometric (LCMS) analysis to isolate environmentally relevant concentrations of emodin from soil and water samples from Carroll University’s Greene Field Station (GFS) in Genesee Depot, Wisconsin. Emodin is a three-ringed, anthraquinone-like compound released from buckthorn (<i>Rhamnus cathartica</i> and <i>Rhamnus frangula</i>), an invasive species ubiquitous throughout the GFS conservancy site. Studies suggest that emodin has negative impacts on organisms like amphibians and amphipods by increasing mortality and malformations and by decreasing body mass. Water and soil samples were collected from areas where the amphibian and amphipod organisms were observed and their biological properties cataloged. Water samples were processed by solid-phase extraction after the extraction efficiencies were optimized. Soil samples were processed by ethanol/acetone extractions followed by centrifugation and filtration. The sample isolates were analyzed by LCMS using a binary mobile phase and standard reversed-phase column. The LCMS method was tested for reproducibility, limit-of-detection, and limit-of-quantification.
