Soluble cereal arabinoxylan, and feruloylated oligosaccharides (FAXOs) generated from it by chemical, enzymatic, and thermal treatments, may promote healthful lower gastro-intestinal functioning beyond traditional benefit from inert dietary fiber. We have isolated FAXOs from rice bran fiber following 1 hour hydrolysis with hot 100 mM trifluoroacetic acid and batch-adsorption to Amberlite XAD-4 resin. We recently reported prebiotic properties for this crude FAXO fraction, finding anaerobic fermentation profiles with human fecal bacteria are comparable to fructo-oligosaccharides (S.-O. Lee et al., submitted to Nutrients). We described separately preliminary evidence for rice bran FAXO activity in modifying epithelial barrier function with T84 colon epithelial cells (B.J. Savary et al., AGFD #4, 252nd ACS National Meeting). We report here structural and compositional features of FAXO sub-fractions separated by C18 solid-phase extraction. Most interestingly, we present evidence that glucan structures covalently coupled to feruloylated arabinose are composed of linear (1α →4) linkages.
