Polarized resonance synchronous spectroscopy (PRS2) is a new optical spectroscopic technique that has enabled the quantification of a series of fundamental material optical parameters that have been totally inaccessible or difficult to obtain before. However, the current data analysis is tedious because the effect of multiple instrument parameters need to be corrected from the as-acquired spectra. Demonstrated herein is a simplification of the PRS2 spectral data analysis that is achieved by explicitly determination of the instrumental response functional (IRF) associated with device used for the PRS2 spectral acquisition. The IRF-corrected PRS2 spectrum is the product of the analyte concentration multiplying its light scattering cross-section spectrum for a nonemitting sample, or multiplying the sum of the sample light scattering cross-section and on-resonance fluorescence (ORF) spectrum. This simplified PRS2 spectrum enables direct visualization of the wavelength dependence of the analyte light scattering and ORF features, facilitating decomposition of the experimental decomposition on the PRS2 spectral into its light scattering and ORF component. The utility of this simplified PRS2 spectral data analysis was demonstrated with a series of representative samples that can be approximated a pure light scatters, pure scatterers, simultaneously photon scatters and scatterers, simultaneous photon absorbers and emitters, all the way to simultaneously photon absorbers, scatters, and on-resonance fluorescence emitters.
