G protein coupled receptors (GPCRs) represent the largest family of membrane-embedded signaling receptors and drug targets. However, these receptors remain difficult to study experimentally and to characterize using structural biology techniques. To address this problem, my laboratory has developed computational techniques to model GPCR structures and their interactions with ligands and downstream protein effectors from structural homologs. Using these models, we were able to rationally design GPCRs with enhanced stability and reprogrammed ligand sensing and signaling functions. Our approach should facilitate the structural and pharmacological characterization of GPCRs, as well as create receptors for interrogating and manipulating cellular signaling.<br/>
