This research is aimed at analyzing volatile organic compounds (VOCs) in mouse urine samples to determine a way to noninvasively test mice to identify those who have breast cancer, those without breast cancer, and efficacy of treatment for breast cancer. Prior studies have demonstrated the use of solid phase microextraction (SPME) and gas chromatography-mass spectrometry (GC-MS) to analyze VOCs in the headspace of urine. SPME adsorbs the VOCs from the headspace onto the fiber and GC-MS separates and identifies the compounds. In order to enable for maximum collection of VOCs, parameters for collection and pretreatment were established. The mouse urine was pretreated with guanidine hydrochloride to denature mouse urinary proteins that are known to sequester VOCs in mouse urine. Urine was also heated, to improve partition of VOCs into urine headspace. Subsequently a SPME fiber was placed into the headspace of vials containing the urine to extract the VOCs present. The SPME fiber was then injected into the GC-MS instrument, and VOCs were separated for analysis. In total, fifty vials of mouse urine were analyzed: 5 with no cancer, 9 with breast cancer, 16 with treated breast cancer, 10 with bone metastasized breast cancer, and 10 with treated bone metastasized breast cancer. Utilizing statistical methods, important VOCs were able to be recognized and the different classes of mice separated through Principle Component Analysis and Linear Discriminant Analysis. Results have demonstrated that there are differences in the VOC profiles of urine of mice with breast cancer, with treated breast cancer, and healthy controls and that SPME/GCMS is a noninvasive way of differentiating between these groups.<br/>
