From our previous studies, modified nucleotides have been detected in DNA from crop plants treated with pesticides. These adducts arise from nucleotide bases directly binding with the pesticide or metabolites or binding with products of lipid peroxidation caused by oxidative stress to the plant. This genotoxic and oxidative stress is likely to be responsible for discoloration of foliage, underdevelopment of the plant, and interference with other plant biochemical systems. Previous work had detected adduct formation from chlorinated aromatic pesticides in <sup>32</sup>P postlabeling studies. In this study two chlorobenzonitrile pesticides, dichlobenil and chlorothalonil, were examined as viable candidates for adduct formation due to their activation toward aromatic substitution by nucleophiles such as DNA bases. In vitro experiments were performed by direct reaction of guanosine with each pesticide under various pH and solvent conditions. Adduct formation was verified with HPLC chromatography.  Chromatographic fractions were analyzed by LC-MS/MS and FT-NMR for structure elucidation of the adducts.  In vivo experiments were carried out by treating crop plants with these pesticides. DNA from treated plants was extracted and digested to mononucleosides, which were then analyzed by LC-MS/MS.<br/>
