Infectious disease remains an imminent danger in developing countries throughout the world but diseases, like malaria, can be better treated and managed if detected early in high-risk areas. Early diagnosis is a challenge since currently available point-of-care test kits for malaria have suffered from poor sensitivity. A sensitive, early detection test could provide low-resourced regions with improved detection tools needed to reduce transmission of disease. Porphyrin nanoparticles have been previously used in signal amplification for biomolecule detection and could potentially provide 10<sup>3</sup>-10<sup>4</sup> times signal amplification per protein target. In this work, anodic stripping voltammetry (ASV) has been employed in the detection of cobalt protoporphyrin IX (CoPPIX) nanoparticles which are then used in a protein sandwich assay. The assay involves a protein target sandwiched between two antibodies conjugated to two different beads or nanoparticles. Magnetic beads (MBs) are used to collect and purify the target while CoPPIX nanoparticles act as a signaling mechanism that can be detected via ASV and correlated to protein concentration.  Detection of CoPPIX had been optimized via ASV with a detection limit of 84 nM. This should translate to protein target detection in the pM range, which is clinically-relevant for targets such as the histidine-rich protein II of malaria.
