Milk thistle (Silybum marianum) is a medicinal plant commonly used as a food supplement for liver protection against hepatitis C virus (HCV). The fruits are very rich in lipids (&gt;22%). In this investigation dry fruits were extracted using CO2 supercritical fluid extraction to obtain the crude lipid components. Lipid extract was fractionated using preparative thin layer chromatography to polar and nonpolar fractions. Triacylglycerides (TAG) were analyzed using Thermo Scientific Ultimate 3000 HPLC LCQ Fleet ion trap LCMS system. Fatty acids were analyzed as their methyl esters (FAMES), unsaponifiable compounds as their TMS derivatives using gas chromatography mass spectrometry (GCMS). Major identified fatty acids were 18:2 (62%), 18:1 (22%) and 16:0 (9%). 5-cholesten-3-β-ol, stigmasterol, β-sitosterol and ergosta-7, 22-dien-3β-ol were the major identified compounds in the unsaponifiable fraction. 21 different TAGs were identified using MSMS3 experiments.
