Hydrogen sulfide (H2S) has recently been recognized as a ubiquitous gaseous signaling molecule in mammals, termed gasotransmitter. Understanding the physiological roles of H2S provides a complex challenge for chemists to synthesize tools that mimic the sustained manner of endogenous H2S production. We report here the synthesis of H2S-releasing polymeric systems based on N-thiocarboxyanhydrides (NTAs) and novel disulfide model small molecule H2S donors. These H2S-releasing functional handles allow for specifically triggered, extended H2S release in aqueous media from the novel water-soluble polymeric H2S donors. These polymeric H2S donor systems also provide a platform for the study of other sulfide species that may be present in vivo, such as carbonyl sulfide (COS) and discrete persulfide species (HS-SR), in addition to H2S. Additionally, these H2S donors have shown antioxidative properties and efficacy in promoting endothelial cell in vitro, helping to further the field of H2S donors towards clinical relevancy.
