Glutathione-S-transferases represents a family of well-studied enzymes responsible for metabolizing the conjugation (combining) of toxic substances needing electrons (electrophile) with the electron-rich biomolecule glutathione (nucleophile) through the GST assay. The accepted standard (control) for this assay is 1-chloro-2, 4-dinitrobenzene (CDNB). To begin understanding the nitro groups positional role in this nucleophilic aromatic substitution reaction, we used three di-substituted chloro-nitrobenzenes with different positional nitro groups ([1,2]-ortho, [1,3]-meta, [1,4]-para). The compounds were added at varying concentration to the GST assay mixture and monitored by UV-VIS spectrophotometer for the appearance of conjugate formation at λmax=340 nm. Our results indicate the possibility of competitive inhibition for ortho and para position while meta-substituted compound trend line was inconclusive. More studies are needed to confirm observed trend line data.
