A novel and facile methodology for the synthesis of α-substituted chalcones is described. The one-pot Knoevenegal reaction has been expanded and optimized to include cyano, fluoro, ester, bromo, chloro and keto chalcones. Previously, attempts to synthesize chloro and bromo chalcones have yielded an epoxide product, but recent developments have incorporated acid catalysis to obtain the desired chalcone. Diversifiaction of the aromatic groups on the chalcones has yielded a broad library of α-substituted chalcones in high yields. The methodology has also been adapted to include α-substitued alkenones in which the library of α-cyano substituted alkenones has expanded to include twenty alkenones.
