The characterization and role of specific post-translational modifications of proteins remains a challenging task. Metabolic chemical reporters (MCRs) of glycosylation are structural analogues of monosaccharides the contain bioorthogonal functional moieties that enable the direct visualization and characterization of glycoproteins from living cells. With MCRs, our lab has probed the role that O-GlcNAcylation, a post-translational modification that involves the single addition of O-GlcNAc to proteins, plays in development of cancer and neurodegeneration. We and others have demonstrated that current MCRs are metabolically transformed and enter various glycoslation pathways —demonstrating the need to develop selective MCRs. We’ll discuss the synthesis, evaluation, and the current progress with our MCRs as chemical probes and their role in illuminating the chemical biology of O-GlcNAcylation.
