Mycobacterium tuberculosis (Mtb) is a common bacterium that causes tuberculosis, an extremely infectious and deadly disease. Due to improper use of antibiotics, multi-drug resistant Mtb has increased in prominence and has the potential to become a global health crisis, with nearly 500,000 cases in 2015 alone according to the World Health Organization. Trehalose is a sugar that is vital to the survival of Mtb, along with other bacteria, plants, and invertebrates; however, it is not present in mammalian cells. In Mtb, trehalose has many functions, but is primarily used as a vital component in the structure of the cell envelope. The enzyme that incorporates trehalose into the cell wall of Mtb, Ag85, is promiscuous with the analogs of trehalose that it will accept, allowing it to incorporate synthetically modified trehalose analogs into the bacterial cell wall. This makes trehalose an interesting target for drug development in the fight against drug resistant<br/>Mtb. The goal of this research focuses on the synthesis of trehalose analogs that are attached to diphenyl porphyrin (DPP), tetraphenyl porphyrin (TPP), or a bacteriochlorin, all of which are photosensitizers that may potentially be used for the photocynamic inactivation of Mtb.
