We have developed a High-Throughput Chemistry Platform (HTCP) capable exploring chemical reaction space with unprecedented breadth. The platform can interrogate reaction partners, catalysts, and reaction conditions using automation in concert with advanced analytical methods and statistical modelling techniques. Up until the start of the 21st century, chemists have carried out reactions in individual flasks, limiting the potential for broad exploration. Leveraging the capabilities of recent advances in automation and analytics, the HTCP operates by carrying out reactions in 384-well and 1536-well plates at volumes of 1-10 mL which enables rapid evaluation of 103 – 105 reactions simultaneously. In this presentation we will describe our efforts in developing the HTCP and its use in exploring the Petasis reaction to demonstrate a transformative approach for the discovery and optimization of chemical reactions.
