Tissue necrosis or oxidative browning of Camellia sinensis var. sinensis explants in tissue culture often results in the failure of callus induction and plantlet regeneration, due to substantial phenolic compounds exuded from the cut sites of tea explants, in which the extraordinarily abundant phenolics are present. The aim of the present study was to examine the effects of 2-aminoindane-2-phosphonic acid (AIP), a competitive inhibitor of phenylalanine ammonia lyase (PAL) catalysing the first committed reaction in phenylpropanoid pathway, on phenolics-induced tissue browning and necrosis. Elimination of the endophytic microbes associated with leaf explants of the field-grown plants was also studied using timentin and gentamycin both supplemented into Gamborg B5 medium with addition of 2 mg/L 6–Benzylaminopurine (BAP), 1 mg/L Thidiazuron (TDZ), 10% coconut water and 300 mg/L L-glutamine. Our results indicated that the inhibition of PAL using AIP combined with the two tested antibiotics could open up new doors to well control oxidative tissue browning and endophyte contamination for in vitro culture system for genetic manipulation in tea.
