A highly sensitive and selective biosensor was developed for the early diagnosis of prostate cancer with the simultaneous quantifications of prostate specific antigen (PSA) and vascular endothelial growth factor (VEGF) in a sample. Modified PSA and VEGF aptamers bound with paramagnetic beads in PBS (100 μl) were mixed with a standard or sample containing PSA and VEGF for 30 min at room temperature. After the incubation, paramagnetic beads were washed 3 times using a magnetic separator. Then, paramagnetic beads were dispersed in deionized water (100 μl). Paramagnetic beads (20 μl) in deionized water were mixed with Tetra-n-propylammonium hydroxide (TPA, 20 μl) in a detection cell. Finally, the light emitted from the detection cell with the addition of 4 mM 3,4,5-trimethoxylphenylglyoxal hydrate (TMPG, 200 μl) was simultaneously detected with two different photomultiplier tubes having different filters. With the increase of PSA concentration in a sample, the strength of the red light observed at 615 nm dropped exponentially. Also, the strength of the green light sensed at 523 nm was enhanced with the increase of VEGF concentration. The limits of detection (LOD) of the biosensor were as low as 0.03 ng/ml for PSA and 0.4 ng/ml for VEGF. Based on the research results, it was confirmed that the biosensor can be applied as an advanced and new medical device capable of simultaneously quantifying PSA and VEGF in a sample for the early diagnosis of prostate cancer with good accuracy, precision, and reproducibility under the acceptable statistical error ranges.
