When hypoxic tissue is rapidly reoxygenated, the sudden shift in cellular metabolism gives rise to a wave of cell death known as reperfusion injury. This condition, which occurs in heart disease, stroke, and organ transplantation, is not readily prevented by any clinically approved drug. In this presentation, we will discuss strategies to develop new therapeutic agents for the prevention of reperfusion injury. Our efforts are focused on exploiting the novel properties of coordination complexes of ruthenium to elicit a cytoprotective effect in reperfused cells. These compounds are designed to inhibit mitochondrial calcium uptake, a phenomenon that is known to give rise to reperfusion injury, and to selectively release hydrogen sulfide, a gasotransmitter with well documented cytoprotective properties. This work highlights the potential of coordination complexes for use in the treatment of reperfusion injury.<br/>
