Cross-coupling reactions continue to become an increasingly utilized class of transformations for the synthesis of complex molecules. Growth in this field stems from the ever-growing list of electrophilic and nucleophilic partners that can be coupled by the use of Pd and Ni catalysts. Recently, strong carbon-oxygen bonds have been recognized as abundant alternatives to more activated carbon-halogen containing starting materials. In our lab, the C(acyl)-O bond of esters has been identified as a viable site for oxidative addition, enabling the direct synthesis of esters, amides, and biaryls. Our progress in this field will be reported, including a discussion of how high throughput tools have been utilized to facilitate the discovery process.
