The epidermal growth factor receptor (EGFR) is overexpressed in a number of epithelial malignancies and its activity enhances tumor growth.  Ligand-induced EGFR oligomerization in the membrane has initiated interest in the function of EGFR oligomerization in signaling. However, the role of receptor distribution in signaling remains poorly understood. The lack in understanding is primarily due to the absence of tools capable of quantifying the impact of EGFR clustering on length scales of tens to hundreds of nanometers. Plasmon coupling between metal nanoparticle (NP) immunolabels targeted at cell surface receptors can be used as an optical transducer of receptor clustering in cancer cell lines. The plasmon coupling imaging approach can test the role of the cortical actin network and protein-lipid interactions in patterning EGFR clustering.
