Global concerns have been raised on tetrabromobisphenol A (TBBPA) and its analogs because of their widespread application and potential health risks. Although previous studies have shown that TBBPA could increase adipogenesis, the effects of emerging TBBPA analogs, with high structural similarities to the parent compound, still remain a blur. In this study, 3T3-L1 adipocytes were treated with TBBPA and its eight analogs during the adipocyte differentiation for 8 days. Their adipogenic effects were evaluated by the mRNA expression of the related transcriptional regulators and adipogenic markers (PPARγ, adiponectin, FABP4, perilipin, C/EBPα, RXRα, and ATGL). The results showed an obvious increase of the mRNA expression induced by TBBPA analogs and the parent compound. The EC50 values of tetrabromobisphenol A bis(2-hydroxyethyl ether) (TBBPA-BHEE), tetrabromobisphenol A mono(2,3-dibromopropyl ether) (TBBPA-MDBPE), and tetrabromobisphenol A mono(allyl ether) (TBBPA-MAE) were lower than those of TBBPA, suggesting the higher potential of these emerging analogs in promoting adipogenesis. These findings would be important for risk assessment on emerging TBBPA analogs.
