There continues to be a need for having a minimally noninvasive glucose sensor for frequent monitoring of glucose levels in diabetics to maintain optimal management of the disease. To address this need, we have devised a two-component system comprising a fluorescent dye and a boronic acid-appended bipyridinium salt molecule that acts dually as a fluorescence quencher and a saccharide receptor. Mechanistically, in the absence of saccharide, a non-fluorescent ground-state complex forms through an ionic association between the dye and the viologen boronic acid, resulting in a greatly reduced fluorescence of the dye. The electrostatic attraction between the dye and the quencher is lost, however, upon the binding of the viologen to the saccharide causing the dye-quencher complex to dissociate. This produces a fluorescence recovery that is directly proportional to the saccharide concentration. These boronic acid-appended bipyridinium salts are tunable receptors and can be designed to increase glucose selectivity over that of other boronate-forming analytes. This two-component system ensemble was immobilized in a hydrogel matrix allowed for the development of an implantable continuous glucose monitor. Additionally, this two-component ensemble has been utilized as a chemical assay for measuring sugar markers of interest in urine for gut permeability assessment.
