Cupredoxin is a protein containing type 1 copper center and generally mediates electron transfer between proteins. Although type 1 cupredoxins including Amicyanin, Plastocyanin and Rusticyanin execute similar function and have structural homogeneity, they exhibit wide range of Em value. For instance, rusticyanin has much higher Em value than other cupredoxins due to serine residue nearby copper center. In this study, Asn47 and Asn54 around copper center of amicyanin were substituted to serine like rusticyanin to analyze effect of serine on amicyanin. As a result, the mutation did not affect protein expression and spectrum. However, it changes redox properties of amicyanin such as redox titration and Em value. This result represented that Em value of copper could be changed by residual property of amino acid nearby copper site.
