Liposome has low toxicity, high stability and solubility because it has the lipid bilayer structure similar to biomembrane. In particular, the stability and shape of the liposome is important when it is used as a delivery vehicle and the osmotic pressure makes an significant effect on the shape of the liposome.<br/>Here, we measured the size and stability of liposomes using AsFIFFF and DLS, and Zeta-potential under various osmotic pressures. As the higher the lipid concentration was and the lower the storage temperature was, the more the size was stable. Moreover, we analyzed and assessed the mean size and the shape change of liposomes under various osmotic conditions. Based on these results, the liposome containing a fluorescence dye, calcein was delivered to HeLa cells under three osmotic conditions to evaluate the effect of the shape change on its delivery efficiency.<br/>Autophagosome is a spherical double layer membrane that enwraps organelles like ER (Endoplasmic Reticulum). After the lysosome is fused into the autophagosome, it degrades abnormal intracellular proteins or organelles for the recycling of biomolecules and diverse physiological processes. Dysfunction of autophagosome is associated with many human diseases like liver diseases, cancers, and some infectious diseases.<br/>In this study, we formulated the liposome containing Alexa488-labeled streptavidin linked with autophagosome-specific peptide chains. For its efficient delivery, we induced the shape change of the liposome by modulating the osmotic pressure. As a result, the optimum peptide sequence for autophagosome detection was screened using the liposome formulated with fluorescent streptavidin-based nanoparticles as well as the effective delivery condition.<br/>
