Pfizer’s Green Chemistry program started in 2002 and, since then, we have striven to incorporate the 12 Principles of Green Chemistry into our processes. More recently, Pfizer has engaged in collaborations with academicians to develop solutions to the challenges that increasingly complex drug candidates pose to develop efficient and scalable synthesis.<br/>The first part of Pfizer’s presentation will describe collaborations with academicians such as Phil Baran (Scripps), Dan Weix (Wisconsin), and Yin-Quan Yu (Scripps) to develop greener alternatives applicable to the development of pharmaceuticals in the areas of non-precious metals such as nickel, as an alternative to palladium, and C-H activation for molecular diversity generation.<br/>The second part of this presentation will comprise examples of application of green technologies to Pfizer’s portfolio. Thus, examples of C-H activation and transition metal catalyzed cross-coupling in water for the synthesis of drug candidates will be discussed. Finally, sertraline, an antidepressant commercialized by Pfizer as Zoloft<sup>TM</sup>, will be showcased as a great example of the application of green chemistry principles to improve a process in terms of waste reduction and the use of green solvents on commercial scale.
