Breast cancer is the most common type of cancer detected in women, but there is no noninvasive method to screen for the disease. Current methods to identify breast cancer require a biopsy, but over half of women recommended for biopsy from screening mammograms, ultrasounds, and other radiographic procedures do not have breast cancer. Volatile organic compounds (VOC’s) include metabolites that can provide information about health and disease.  We hypothesized that mice with induced breast cancer would express different VOC’s than healthy mice and that mice with mammary pad confined breast cancer would express different VOC’s than mice with breast cancer metastasized to bone. We therefore aimed to classify different types of cancerous and healthy mice via differences in VOC’s in mouse urine. To study this, 4T1.2 murine breast cancer cells were injected into BALB/c mice either in the mammary pad or into the iliac artery. After 18 days, urine was collected, VOC’s from urine headspace were concentrated by solid phase microextraction (SPME) and data were analyzed by gas chromatography-mass spectrometry quadrupole time-of-flight (GC-MS QTOF) using an Agilent 7980A/7200 Accurate-Mass Quadrupole Time-of-Flight GC/MS System. Univariate and multivariate statistical analysis of the data was undertaken using Wilcoxon’s rank-sum, the Student’s t-test (p-value&lt;.05), linear discriminant analysis (LDA) and leave one out cross validation (LOOCV). Results show that there is a difference in VOC composition between mice that do not have cancer and mice that do have cancer. VOC composition in urine also varies between mice that have mammary pad cancer and bone metastatic cancer.
