The experimental quantification of enzyme activity exhibits large variance in the scientific literature. In this talk, we present our analysis revealing two leading causes of the large variance reported in estimates of enzyme kinetic parameters. The first source of variance is the limited adoption of Standards for Reporting Enzymology Data (STREDA). Better reporting of enzyme kinetic experiment protocols is required for improving reproducibility and making it easier for newcomers to enter the field. The second culprit is the lack of standardized protocols for experimentally measuring, and mathematically estimating kinetic parameters. Our analysis of current experimental design protocols used to estimate Michaelis constants of enzyme catalyzed reactions show that reported values can exhibit variations of four order of magnitudes. Better reporting guidelines to measure and estimate kinetic parameters will lead to accurate measurements, and will enable advances in the biochemical and pharmacological sciences.
